Prof Mario Suva is Assistant Professor of Pathology, Harvard Medical School Assistant Molecular Pathologist, Massachusetts General Hospital Institute Member, the Broad Institute of MIT and Harvard .

Abstract

Human gliomas are incurable malignancies. One of the daunting challenges is repre-sented by their important level of hetero-geneity. In this lecture I will show how one can leverage cutting-edge single-cell genomic tools to interrogate human gliomas in adults and children. I will review the lessons learned in IDH-mutant oligoden-droglioma, IDH-mutant astrocytoma, H3K27M-mutant pediatric midline glioma and IDH-wildtype glioblastoma. I will show how single-cell genomics have shed light on cellular lineages of malignant cells, cancer stem cell programs, linking expression and genetics. I will also discuss lessons learned in the composi-tion of the tumor microenvironment, both myeloid cells and T cells. Finally, I will demonstrate how integrating single-cell genomics with large bulk profiles such as those from the TCGA can help shed light on brain tumor classification.
