Metagenomic researches of viral communities (viromes) are currently one of the most dynamically developing approaches for the detection of rare, mutant and discoveries of previously unknown viruses. High-throughput sequencing methods are widely used in such researches. An important factor limiting metagenomic researches of viruses is a presence in analyzed samples cell-free eukaryotic and prokaryotic nucleic acids (NA), the size of which is much more than viral NA. The known methods for non-viral NA depletion or viral particles enrichment (concentration) require a relatively large amount of input material, are insufficiently universal and not suitable for samples containing partially degraded virions. Our method does not have these disadvantages and allows the universal purification from free NA for samples potentially containing viral particles at least 4 times.
