Hyphenation of fast liquid chromatography with high-resolution mass spectrometry for quantitative proteomics analyses.

Targeted analyses performed on high-resolution accurate mass spectrometers hyphenated to liquid chromatographs have opened new opportunities in quantitative proteomics. A new extension of the parallel reaction monitoring (PRM) techniques uses the internal standards added to samples to dynamically control the data acquisition and maximize the analysis of the endogenous peptides. This account describes the hyphenation of ultra-HPLC with a high acquisition rate quadrupole-orbitrap instruments to properly sample the sharp elution profiles of UHPLC separations, possible with the IS-PRM technique.
